Synthetic oligonucleotides are indispensable in many different fields. Millions of oligonucleotides are synthesized every day for use in research laboratories, hospitals, and industries. With the development of chemical modifications of oligonucleotides, applying antisense oligonucleotide-based strategies for the treatment of diseases has become possible. Libraries of vast numbers of oligonucleotide aptamers, microarrays of thousands of oligonucleotides on surfaces, and building block sequences for DNA nanostructures are also prepared by chemical synthesis. Common for all the oligonucleotides described above is that they are synthesized from nucleoside phosphoramidite building blocks[1].

High-quality phosphoramide and a good solid carrier are important factors for the successful synthesis of long oligos with artificially modified backbones, which could not be prepared easily by other methods.

The artificial analogs include alkyl/arylphosphonates, phosphotriesters, phosphoramidates, phosphorothioates, phosphorodithioates, phosphoroselenates and boranophosphates, which are attractive as antisense molecules. The phosphoramidite strategy has been and is being most widely employed in both liquid-phase and solid-phase syntheses of various kinds of oligos[2].
