Design and Chemical Modification of CpG Oligonucleotide

CpG stands for nucleotide pair, in which G follows C in the DNA strand. CpG dinucleotide is unevenly distributed in the human genome, while CpG remains or exceeds the normal probability in certain regions of the genome, which is called CpG island. Modification of pentose phosphate skeleton structure in the CpG - DNA skeleton structure is related to its immune adjuvant effects. The physical properties, including helical stability, melting point, resistance to nuclease degradation and uptake of CpG motif by cells have changed. The immune activity is significantly enhanced. CpG ODN can be divided into K and D two types, according to the difference of immune stimulation effect of CpG ODN on human peripheral blood mononuclear cells. They have different skeleton structures. K- ODN is modified by diester total thiophosphate and consists of multiple TCGTT and P or TCGTA motifs. D- ODN is a mixed skeleton of diester phosphate bond (PO) P dithiophosphate bond (PTO). It centers on the motif of purine - pyrimidine - CG - purine – pyrimidine and has a certain flanking sequence, forming a complementary palindromic structure. The 3 'end and 5' end is added poly G tail. Research on the different roles of the two types of CpG ODN has become one of the hot spots in the research field of CpG ODN.

  • Mouse immune adjuvant. The component of mouse immune adjuvant is synthetic oligodeoxynucleotides (ODN) with non-methylated CpG motif (CpG ODN) as the core. CpG ODN adjuvant is optimized to specifically activate the immune response in mice/rats and improve the titer of antibodies. Bacterial DNA activates innate immunity by interacting with TLR9. Its fragment, a synthetic CpG ODN, can also act as an adjuvant. The use of correct CpG motif can trigger B cell activation and directly or indirectly induce the production of Th1 and proinflammatory cytokines, which ultimately realize the maturation and activation of antigen presenting cells (APC).
    • It can help clients save immunogens and adjuvants. The amount of immunogen used in mouse immune adjuvant is less than that of traditional freund's adjuvant.
    • It can save time for customers. Unlike the complex emulsification process of freund's adjuvant, the antigen and adjuvant can be simply mixed to immunize animals. A strong immune response can be achieved within 35 days after immunization.
    • It doesn't destroy the natural conformation of antigens. It is easily to screen and obtain monoclonal antibodies against a conformational antigen epitope.
  • Chromed CpG bone marrow cell culture additive. It is composed of CpG ODN DSP30 and rH-IL-2, which can effectively stimulate the division of human B lymphocyte. It is mainly used in the culture of bone marrow or peripheral blood B lymphocytes with chronic lymphocytic leukemia (CLL). It can also be applied to the cell culture of chronic B cell lymphoproliferative syndrome. It can perform cytogenetic analysis and improve the detection rate of chromosomal abnormalities.
    • This product is easy to use and add. It can aim at CLL specimen.
    • CpG ODN can effectively prevent the degradation of ODN by intracellular DNA enzymes after complete thioylation.
