Design and Chemical Modification of miRNA and Anti-miRNA Oligo

MicroRNA (miRNA) is single-stranded RNA molecules with endogenous length of 18-22nt, which plays an important role in controlling gene expression after transcription. From embryonic development to cell apoptosis and tumor growth, miRNA is one of the most important factors in the process. The complex and detailed regulatory mechanisms of miRNA make the entire signaling pathway a promising therapeutic drug (usually in the form of miRNA analogs) or a therapeutic target (usually in the form of anti-miRNAs) in many pathological processes. When AMO is introduced into cells or in vivo, it is specifically bound with miRNA to form AMO-miRNA hybridization, preventing miRNA from binding to target mRNA and activating RNaseH to degrade miRNA. Common ways of chemical modification include phosphoro-thioate backbone, 2'-O-methyl, 2'-O-methoxyethyl, 2'- flouro, locked nucleic acid and peptide nucleic acid.
