In Vivo Activity Evaluation of Oligonucleotide Drugs

In vivo activity study is an important link in the research and development of new drugs. We provide but are not limited to:

  1. Hollow fiber assay. Hollow fiber assay is a rapid in vivo analysis technique that can be used to evaluate the cytotoxic effects of drugs. Human tumor cells can be encapsulated and cultured in hollow fibers and implanted subcutaneously or intraperitoneal in nude mice to evaluate the efficacy of the compounds under test.
    • Hollow fiber assay can evaluate the efficacy of antitumor drugs against multiple different cell lines at two different sites in the same animal.
    • Hollow fiber assay uses a semi-permeable fibrous membrane with good biocompatibility. The lumen of the fiber can encapsulate tumor cells, which is suitable for tumor cells with different tissue origin and cellular characteristics.
    • For drugs with repeatable activity that have been identified in in vitro antitumor screening, this can be used as a preliminary in vivo evaluation to determine the potential activity of the drug.
    • Hollow fiber assay helps customers to select compounds that can be prioritized for further screening of transplanted tumor models, significantly reducing the amounts of active compounds required for direct graft detection.
  2. Transplanted tumor model. This model is widely used in the research and development of anti-tumor drugs, and is an important link in the research and development of anti-tumor drugs. The results comprehensively reflect the in vivo effect of drugs and provide important data for evaluating the drug ability.
    • The accelerated test includes the trial of six months in one environment.
    • Three batches of samples are taken for the accelerated test. It is recommended to carry out the accelerated test at a temperature at least 15 ° C higher than that of the long-term test. The experiment can be carried out under the conditions of 40 ° C±2 ° C and RH75% ± 5% for 6 months.
  3. Efficacy test of anti-cerebral ischemia drugs. This is a common and reliable method for screening stroke drugs.
    • The candidate drug is prepared into injection and administered by injection after reperfusion in rats.
    • Rat behavior is detected at 24 hours and 48 hours after administration. After 48 hours behavior detection, brain sections are quickly stained and ischemic area is calculated. Compared with the model group, if the rats injected with the candidate drug show improvement in behavioral score or cerebral ischemic area, it suggests that the candidate drug may have potential effect on stroke.
